Try running calibre in Windows Safe Mode Safe, if that works it's some 3rd party program that runs at startup getting in the way, probably an AV or another 'security' gadget.

Try restarting with no optional plugins, there's an option in the Preferences drop down menu

Use Task Manager to see what child processes calibre has running, click the right pointing arrow, viz:
