This isnt plugin related, according to that debug log something is interfering with calibre before it even gets to the stage of creating its UI long before plugins are even loaded. I am very surprised that this would occur in Safe mode as this is almost certainly some program on the computer interfering with calibre. The log indicates the QApplication object was constructed and then it has hung.
Try temporarily disabling the antivirus and also trya clean reinstall that is un-install, delete c:\program files\calibre2, then re-install (this wont affect your books/settings)
