So I have no idea what is going wrong - the fact that the ebook is already in Calibre in its converted version maybe? (but it seems a remote hypothesis). Does anyone have a suggestion ? (I certainly hope so...)

Did you export the keys for DeDRM from the old install and import them in the new install? What version of KindleforPC are you using? The new version won't work. You need an older version.
