PDF format uses a fixed-layout that often does not work well with calibre conversion. Calibre will convert the PDF into a reflowable EPUB but the result may have a variety of flaws. See Read this before Posting PDF Questions for details.

The error that is produced in this case, "KPF file is missing", results from the Kindle Previewer crashing when attempting to convert the reflowable EPUB produced by calibre into KPF format. This can happen for extremely large and complex books. I suspect that is the case here.

I recommend that you convert your PDF to AZW3 format in calibre and then check whether or not the result is readable using calibre's viewer. If so then that format will probably be a better choice than KFX for this particular book.
