No, you do not need to format the µSD card before copying the image to it. The image has a chunk of free space, two Linux ext4 partition and a FAT32 partition which will be created when the image is copied to the card.

I prefer to use a new µSD card when copying the image since all too often the original µSD cards has problems which caused the need for a new image. You can try using the SD Memory Card Formatter for Windows/Mac from the SD Association before re-imaging the old µSD card.
