Which version of Windows and is calibre updated to the latest version? As of today, the current version is 6.28.1. If you are running WIndows 10 64-bit, it is full updated? An older version of Windows 10 will cause calibre not to run.

Stop Avast from running and then try to run calibre. If that doesn't work, boot into safe mode and try calibre again. If safe mode works, stop everything that's starts on startup and add them back in one-by-one until you find what's causing the problem.
