As @theducks said, you need a second connection after the books have been imported to send the metadata. The synopsis is taken from the Comments visible in the Book Details pane in calibre.

You also need to have calibre set to automatic metadata management (check under Sending books to device in calibre preferences) and in the KoboTouch or KoboTouchExtended driver configuration, you need to have Update metadata on the device and Update metadata on Book Details pages checked. See the attached images. You can disregard the Subtitle and Book Stats entries.
