I have the latest and greatest version of calibre installed on Windows 10 x64 with 8000+ titles in the library. When enabled, pressing a key in the library view will search for the next cell in the current column where the value starts with the letter pressed. If there is no such cell then nothing happens. If the letter is a shortcut then the shortcut takes precedence.

If you don't like the last sentence, change the single character shortcuts. To find those shortcuts; JobSpy has a tool to export the shortcuts into a CSV which you can sort.
