That probably means that a 3rd party program that is run at startup is preventing calibre from running; likely candidates include anti-malware, shell extensions, etc.

To determine which one, use a process of elimination (e.g a binary chop) via Task Manager's->Startup->Disable function.

Oh, and have look at the FAQ, there are several 3rd party programs that are known cause problems if run with calibre ==>> Frequently Asked Questions
