In addition to what Quoth wrote there are many books for which the Download & Transfer option will not be available. Some possibilities are books in Arabic or Indian languages, books borrowed from Kindle Unlimited, free book samples, many textbooks, and many English language books published in India.

If you have an e-ink kindle registered to your Amazon account and are not seeing the Download & Transfer option for a particular book you can post a link to the Amazon listing for the book and someone can look at it to see if it falls into a non-downloadable category.
