Today the 'Kindle for PC' App refused to download a purchased ebook unless I update the Kindle App. After I did the download worked but Calibre could no longer import the file.
Can anybody tell me what to do?

Kindle for PC 1.17 uses AZW, which DeDRM could break. The newest version of Kindle for PC uses a newer format called KFX, which is (except a few early versions) not breakable.
Fortunately, this only currently affects books published after January 3. I assume you have one.
If you have an e-Ink Kindle, you be able to use "download and transfer" to get a breakable format. If you don't and are not able/willing to get one there isn't much option other than seeing if book is available elsewhere. (Alas, if it's a self-published author on Kindle Unlimited, you can safely assume they're using KDP Select and exclusive.)
