Update DeDRM before you update Calibre. You want to install 10.0.3 from https://github.com/noDRM/DeDRM_tools

Then install the latest Calibre 6 and update the plugins when prompted for new versions.

What you can do is have a look at all the plugins you have installed and go to the appropriate thread in the Calibre Plugins forum and double check what works with Calibre 6.
