This may be stating the obvious but the error message says it's having trouble writing to directory
C:\Users\User\Documents\eBooks\Calibre database backup.
Have you checked that this directory actually exists on your new PC?
If it's not that then the next thing I'd consider is choosing a backup directory which doesn't contain spaces in its path.
