Were you running the 32bit version of calibre before? There have been some issues with the change-over from 32bit to 64bit. Sometimes it isn't done cleanly and the wrong executable or library gets used. If so, can you uninstall calibre and look to see if there are any 32bit version still shown in the Add/Remove programs and uninstall that. Then remove the "calibre2" directories from both "C:\Program Files" and "C:\Program Files(x86)". Then reinstall calibre.

Otherwise, please run calibre in debug mode and post the log. There might be something failing earlier that is triggering this.
