it may help to turn on show full kindle previewer conversions logs , I also like to click on create approximate page numbers and/or include kindle preview quality reports, in the settings for KFX Output in calibre.
but for this question, I would say it works best with epub to kfx, because the kfx output plugin or kindle previewer will take say if you convert a mobi to kfx, it will first convert mobi to epub anyway before it outputs it to a kfx file, so no matter what you are working with say azw3 goes to epub before its converted into a kfx file, that's just how the kfx output software in calibre works, I think thats also how kindle previewer does it, first into a epub format then it goes into a kfx container file format.
I hope I helped explain its processing of epub to kfx files, no matter the source you work from in calibre it can be even a pdf to epub then kfx I think that best explains this plugin or kindle previewer.
