Calibre has not used python 2 for at least a couple of years. It could be by a plugin that has not been updated. Post the details of the error message for us to look at.
A possibility is that you have two calibre versions installed and they are conflicting. I managed to do that when I changed from 32bit calibre to 64bit calibre. The 32bit version wasn't uninstalled properly and sometime bits of it would be picked up and cause some very strange errors.
