Calibre should already be doing that. The default configuration will split a name at either of the words "and" or "with" or at an ampersand. This can be changed with the tweak "Splitting multiple author names". If you have changed that in the past, you can reset it in the configuration.
And I did check. I changed the author of a test book to "Peter Miller and Paul Shuster" and when I saved this, calibre showed the author as "Peter Miller & Paul Shuster".
For the current problems, @BetterRed's suggestion is the way to go.

Honestly, don't. If you are doing this for sorting, then let calibre do it. Calibre generates a "sort author" for use in the library. This defaults to "LN, FN". When you export the book, both are put into the metadata and different tools will use one or both of them. Changing the author like that tends to have flow-on effects that are a pain. If you want to display the sort author instead, you can create a custom column and hide the author column.
