Something is blocking calibre's ability to access the python-lib-bypy.frozen file. Perhaps your anti-virus/anti-malware is blocking access to the file? Perhaps the file has been deleted or damaged? Have you looked in your Calibre2\app\bin directory to see if the file is there? This has nothing to do with the epub itself.

More useful information would be the version of calibre, the version of your operating system, any anti-malware/anti-virus programs that are installed, etc.
