If you are referring to booting into safe mode, that is hardly drastic and is often used to test a program with a minimum of Microsoft's bit and bobs running. It is a bit more a pain to get into under Windows 10 but not that hard. Type msconfig into the search box, click on the Boot tab, select Safe boot with the minimal radio button selected, click ok and then restart. To get out of safe mode, same procedure but Normal startup on the General tab.

If calibre then runs, you can start checking your startup items to see which one is interfering with calibre. If calibre doesn't run, then you have some serious issues with your setup.
