Just about every ereader app I have tried on my Poke 2 has a nightmode but, as you say, the in-built NeoReader does not! (I have also found that even the smallest font size in NeoReader is too big for my taste.)
With some of the reader apps, I found that it was necessary to delve a bit into the settings to get them ideally set for e-ink (a lot default to various colours for things such as title, links etc which do not look good on e-ink).
As for Koreader, strangely switching that to night mode causes issues on my Poke 2 - the reader becomes slow, temperamental and subject to freezing. I am not sure why.
