In other words, do we avoid things in the manual, or just leave a one line summary and a link? There may be a few "trick" like things in the calibre manual mixed in with other sections that are not well indexed, like, how to accomplish a specific task using a feature in that section of the manual, and not indexed.
Searching the wiki, looks like there might be half a dozen existing tricks scattered about we could start with.
I know there's been a few questions here recently that were not covered well in the manual. When I have time (if someone else doesn't get there first) I might go through old threads and pick some up.
The only other question would be how to structure it initially.
This can be emergent and restructured later, but wiki pages tend to work better if there's a good initial structure. But it's more important to get started than to get a good initial structure, so...

1. I would say overlap with the manual should be minimal. IF what is in the manual is indexed and all in one place. I agree with your suggestion , all we need is "a one line summary and a link" .
2.Great that will help collecting hints and tips quicker.
3. If you do not have the time to go through the old threads just give me a hint and I will start on them if no one else does.
4. I have not played with Wikis so I have no knowledge of them. I will leave setting the Wiki up to to you or some other member with Wiki knowledge, who would like to help.
5. I have a couple of Hints and tips I will start writing up in the meantime.
