What version of calibre are you running? Calibre 5 breaks the DeDRM plugin. Are you always using the same Kindle serial number for your download or have you entered the serial numbers for both your Kindles in calibre? Download and transfer via USB locks the downloaded book to the serial number of the Kindle you specify in the download.
