My Vista machine got hosed a long time ago and wouldn't launch any .msi installers from Windows file explorer. I had to run them (calibre included) from an administrator command-line with something like:

msiexec /i "path\to\setup.msi"

Worth a shot. And as mentioned by others; make sure you don't need the 32-bit installer. 64-bit Vista machines were a thing, but they were fairly rare birds.
