I don't know the straight way how to add a new book with filename automatically put into comments. So how I do is go to 'Preferences'>'Import/export'>'Adding books' in 'Reading metadata' tab, uncheck 'Read metadata from file contents rather than filename' and use '(?P<comments>.+)' in 'Regular expression'. After adding a new book, everything okay as expected; the filename has been put to both 'Comments' and 'Title' of the book (which I think 'Title' also got filename for failsafe of Calibre not creating a book without title). And when I click 'Set metadata for the book from the selected format', all other fields have been filled with metadata read from the file correctly, including the new 'Title'. The problem is, the 'Comments' has been replaced with new filename from the Calibre library also. I'm okay if it should be replaced with real "Comments" from the metadata in the file. But in case there are no comments metadata, should it be replaced with the new filename again?
