I think the font(s) used in the Kobo GUI are dependent on which locale you have set. For English locale the default GUI uses 2 of the builtin fonts. Georgia is used wherever the firmware asks for a serif font and 'Avenir Next' when sans-serif is specified.

As for the rest of the OP's question I don't know about kerning and font spacing because I'm not sure how I'd test them, but I am pretty sure ligatures (e.g. 'fi') will be used in the GUI if the Georgia/Avenir fonts actually contain those ligatures.

I've updated my own Forma to use Bookerly/Trebuchet throughout the GUI rather than Georgia/Avenir. I can see that the 'fi' ligature is being used when displaying Author 'Gianrico Carofiglio' in both main Book list and Author list.
