When you put the book on the device and ejected, did the importing message get shown? If it didn't, then it means the device didn't see the book. Which usually means it was put in a directory that the device doesn't scan. With older firmware, that meant a directory whose name started with a dot. Now, these are scanned, but, you can add an option to the config file to ignore some directories.
If the import was shown but the book isn't seen, it is usually an error in the book. It could be the structure, something in the OPF or the ToC. I'd need to see the book be able to tell guess why it failed. But, if you use the error checking in the calibre editor and it shows errors, fixing those before resending the book will probably solve the problem.
The other question is when does the tick show in calibre. It should show when you first send the book. But, does it show after disconnecting and reconnecting the device? If it does, then the book has been imported into the devices database. The KoboTouch driver does not scan the device for books, it reads the database. Only books that have been imported by the device will be seen when calibre sees the device.
And for the record, the Kobo devices have absolutely no problem with multiple copies of the same book. If there are multiple files, they will just be added as extra books with identical metadata. I do this frequently for testing purposes. And almost as often by accident.

Yes, when I did the “Send to device” thing there was a brief progress bar on the Glo screen (I assume showing the transfer of the book). The green tick showed after I did the above - however, on ejecting the Glo then reconnecting, the green tick was gone (it was still there for the other books on the Glo). Thankfully, I can now continue with this series of books I’m reading!
