You are running a 32-bit version of KindleGen. Catalina won't run 32-bit apps, requires 64-bit.
You can extract a 64-bit KindleGen from
Kindle Previewer by using right-click to Show Package Contents for Kindle Previewer > Contents > MacOS > lib > fc > bin > kindlegen. Make a copy of kindlegen and move it to your Applications to replace the 32-bit version you're currently using.
