Did you update Calibre recently? Sometimes a new version of Calibre gets flagged by a 3rd party AV software and you can't do anything until you clear Calibre in that program.
Edited to add:
Sometimes it not even 3rd party but the bleeding stuff provided by MS, Defender I think it's called now?
