update. I re read the whole saga.
I confess I thought we were talking about a calibre library on a USB stick but in one post the OP says its a multi-terrabyte drive. ( if that's a self powered one then previous speculation about getting enough current from the USB port for sustained RW stays on the usual suspects list)
I also know that when I have moved fixed drives to new PCs I have encountered ownership issues. windows thinks the drive is "owned" by the other PC so it blocks some operations when attempted on the new PC. the easy fix for that is a take ownership utility or a registry hack to get full ownership of that drive for the current user on the current PC
all the other ( ignored?) troubleshooting suggestions of trying various permutations of different PC, different port, different usb drive remain valid and will help , if OP can be bothered. ( & if he can't its not going to get solved )
