What you are saying sounds like it should have worked. Or, at least I assume you have opened the book in the editor and replaced the existing cover.jpeg with your new one. But, where doesn't it show? Are you saving and opening the book in the calibre viewer or some other ebook reader app or device? Does it show correctly in the preview in the editor? If you are looking at the cover calibre shows in the library, you won't have been updated. You need to set the cover from the book in the metadata editor.
