It could be Onedrive synching.

Recent versions of Windows 10 automatically synchronise the %USERRHOME%\Documents folder to OneDrive, for that reason since calibre version 3.30 the location of the default library changed from %USERRHOME%\Documents\Calibre Library to ???? - I'm not sure. This only affects new installations of course. Synching involves file locking and VSS trickery.

IIRC you could exclude the Calibre Library folder in Documents from OneDrive synching; or you could pause Onedrive synching when you're using calibre; or you could move your library to a location that Onedrive won't synch automatically - such as %USERRHOME%\Calibre\Library. I would opt for the third option

Move the library when calibre is not running, then when you start calibre it should ask where its library went and offer a dialogue to select the new location.
