Have been using Windows Defender AntiVirus and surprisingly that must have been the problem, when I turned it off Calibre worked perfectly. I would have to guess that something in the latest Defender update must have caused the problem since it was working fine yesterday,

Right click the calibre icon in the calibre toolbar and select Library maintenance->Check Library--fix any problems found.
If that does not work
then make a copy of your library then Right click the calibre icon in the calibre toolbar and select Library maintenance->Restore database.
