Where did you input that new template?

1) Calibre > Preferences > Sending books to devices > Save template

2) Calibre>Preferences > Plugins > Device interface plugins > Kindle 2,3,4... Device Interface > "Customize plugin" button > Save template


Because #2 will override #1.
Shorter way of opening that "device interface window" is available when you Kindle is connected to you PC and recognized/detected by Calibre:

Click on "Device" icon > Configure this device
