Is there a way to change/update the source file that was used to generate the ebook in Calibre? In my case the source files for the books are .docx

You'll need to edit the DOCX with a word processor (MS Word or XXXXX Office Writer). By default when you View a DOCX the OS default handler (typically Word or Writer) should open the file.
However, if you check DOCX in
Preferences->Behaviour->Use internal viewer for: list, calibre will do an on-the-fly conversion to a temporary EPUB and hand that off the calibre viewer. I would leave it unchecked and do a DOCX to EPUB conversion and use the
View specific format option to see what the EPUB version looks like.
If you don't have Word or Writer you won't be able to edit the original source, so you'll have to import it into the Editor and save it as an EPUB or AZW3 (or convert first). Once you start making substantive edits to the book's contents in the EPUB/AZW3, the original DOCX source is only worth keeping for historical purposes.
If you use your word processor's Styles facility you should be able to get a nice looking EPUB with the default conversion settings - but if you use your word processor as if it were a typewriter you probably won't.
BTW: Libre Office, which includes Writer, is free if you need it, MS Word of course isn't free.
