I've deleted and reinstalled the latest version of calibre. I've run ccleaner to cleanup registry issues.
@vanbode - try disabling your AV, if that doesn't work try running calibre in Windows Safe Mode.

@TomSpot - if you tell us which AV, we could update the FAQ
