I whitelisted both the Calibre folder and the library folder in my ESET NOD32 anti-virus, and continued to receive the error.

I then whitelisted Calibre in the HitmanPro anti-malware, and still received the error.

I then disabled the HitmanPro anti-malware entirely, and it worked.

So, for anyone else that uses the HitmanPro anti-malware, it needs to be entirely disabled to edit the metadata in Calibre.
