If you sideload from Calibre, your book won't sync with Amazon Cloud. You have to convert the book to mobi or dual mobi, then send it to your Kindle's e-mail address or use the Send to Kindle app (
https://www.amazon.com/gp/sendtokindle). Note that Amazon won't accept azw3, so converting to dual mobi is the best option.
