If you have WiFi/4G on, my experience is that you won't notice any great increase in power consumption while reading. You will, however, then get a sizeable power drain while the Kindle is asleep that doesn't happen if you leave it in aeroplane mode.
So if you read in long sessions, having WiFi on won't make that much difference to battery life, but if you read for only a few minutes a day, leaving WiFi on could result in a dramatic decrease in battery life.
