As DNSB mentioned, you should really reinstall calibre. Download the actual installation from the official site

For the following action take care you user account have admin rights.
Uninstall calibre via control panel ->programs, boot your system (usually not needed but just in case...), make a new installation and allow all activities for calibre if there is a request from your security software.

By the way, there was going something with what DNSB mentioned to you. The command "where" should give you a result, as there is an active installation. Maybe a typo or a restricted user account. You can check this if you like. Copy this one in your cmd window with admin rights and hit enter:
where /F /T calibre*.*
This should show you all files with time stamp and locations starting with the name calibre (usually 8 files).
