Kovid Goyal has a section in the FAQ about it. It is not limited to Windows Defender (which also does detect viruses and malware advising against installation).

Please report the false identification to whatever company you buy your antivirus software from. If the antivirus program is preventing you from downloading/installing calibre, disable it temporarily, install calibre and then re-enable it.
