Regarding to my running from source I got now with version 2.99.8, first run a slightly different error message. Maybe this help a bit more

Spoiler:
Code:
calibre, version 2.99.8
ERROR: Failed to start Content server: Could not start the Content server. Error:

Failed to write to file: C:\Program Files\Calibre2\app\resources\rapydscript\lib/gettext.pyj-cached with error: [Errno 13] Permission denied: u'C:\\Program Files\\Calibre2\\app\\resources\\rapydscript\\lib\\_gettext.pyj-cached'

and a lot of errors in debug mode.
The problem is not with Calibre. The problem is where you installed Calibre. Calibre does not have permission to write to Program Files unless you run it in admin mode. Uninstall Calibre and resinstall to a directory not inside Program Files or Program Files (x86).
