Mostly you don't, the change would not persist.
During the normal boot process, both user areas (p3 and p4) are checked and if found invalid (ext3 and fat-32, respectively), reformatted.
Later in the boot process, the fuse file system layer on mmcblk0p4 is started, and it expects an underlying fat-32 storage.
There are quite a few system dependencies on p4 being fat-32 that you would have to change if you expect the Kindle to run after you re-formatted it.
