Can you check the messages to make sure it is using the correct path? The line starting "Copying file" should show two paths and they will start with the temporary directory that is being used. Once you have added the environment variable and saved it, the next time you start calibre it should use it. If it doesn't, then check the spelling and maybe reboot before trying calibre again.
