With my JetBook Lite I hold it in my left hand. This positions my thumb over the page advance "slider" and my fingers over the battery bulge.
My next favorite ereader is my Nook Simple Touch. I'm still most likely to hold it in my left hand -- probably out of habit -- but occasionally I hold it in my right. There are page advance buttons on both sides. I prefer buttons.
