In iOS 9 I found it depends. If the screen is dark (off) then I have to push the button and then I could put my thumb on it. However, if it was already lit, showing the notification screen, then I could just touch it. I also found that it would insist on my entering my passcode from time to time as well and not accept my fingerprint at all. This was on my iPad Air2 with a magnetic cover. In iOS 10 I am still figuring out all the nuances.
