Depends on whether the host device supports SDXC. SDHC is restricted to max. 32GB, so 64GB cards are made to SDXC specification.
SDXC specification also requires exFAT file system and the card reader manufacturer decides whether they want to enforce this in the firmware (so some host devices may not recognize the card if it's formatted to another file system).
Some older card readers made before introduction of SDXC can be firmware-upgraded to support SDXC, but supporting SDHC specification (and capacities) doesn't guarantee SDXC support.
If the device was made after 2009 it's likely that SDXC support is explicitly mentioned.
