Spotify on the Echo requires a paid premium account. The free version won't play.
As to TuneIn, that happens to me every now and then. One day I'll say "Echo, play KZOK", then another day I'll have to say "Echo, play KZOK on TuneIn". I chalk it up to Alexa being a bit temperamental and out of sorts.
