@m.s - not directly, you could write a Viewer plugin to do what you want, or you could try this.

Copy the calibre shortcut from the desktop to the Windows Send to Folder, if there isn't a shortcut on the desktop, copy from Start Menu or create one in the Send To folder in the usual way.
  • click the Open File icon in the viewer, that will open a file open dialogue,
  • navigate to the book's location, eg in %USERPROFILE%\Downloads,
  • right click the book you want to add, select Send to->Calibre
  • hit escape to get out of the file open dialogue - continue reading
If you have multiple libraries, best to make sure calibre is running with the target library open before you do the Send to
