Calibre will not "see" the Fire if some other software has it open. Are you running anything that watches for MTP devices, perhaps a music or file transfer program? Some people say that Windows Media Player will open MTP devices. Is it running?

If you start the desktop machine in "safe mode", can calibre see the Fire?
