My EPUB3 looks perfect on Readium, exactly how I want it to look, it is validated using ePubCheck 3.0.1 but when I pass it through Kindle Previewer I get one page at a time instead of page spread.

Do you mind looking at the code perhaps to see what mind cause that?
Odedta:

Sorry, sweetie--I don't have to. Look, that is. The coding for FXL MOBI is not the same as the coding for FXL ePUB. How it looks as an ePUB on Readium, etc., is utterly irrelevant.

Yank out the latest KP Guidelines, and check the section on FXL. Is your meta correct? The identifier? Did you lock the orientation? Got your CSS reset applied to each page? Got one CSS sheet per (HTML) page? Got your divs set for each half of each html page?

Really, if you just check the Guidelines (2015.4, I think is the most recent) against the ePUB, you'll likely find the culprit. If memory serves...I'm not 100% sure, but the last I recall, it was virtually impossible to make a FXL MOBI/Kindlebook from INDD, even using INDD-->.ePUB-->KG-->MOBI.

And: what's up with the audio files? I take it you're not selling this via Amazon, or, you have a client that is a "real" publisher, with FTP access for a book with multimedia?

(You know how to find me. Ping if you can't sort it.)
